Probiotics
WonderBiotics Probiotics for Gut Health centers its bloating-and-comfort positioning on B. lactis HN019, paired with a multi-enzyme blend and 11 other strains at 60 billion CFU total. Across three human trials, HN019 has shown positive results on gut transit time and bowel-movement frequency, with the clearest benefit in people who started with three or fewer bowel movements a week17,18. The most recent trial, in 229 adults over 8 weeks, didn't find a significant difference on its main bowel-frequency measure, but people taking HN019 reported less bloating and abdominal pain than the placebo group19. These three trials were funded by the ingredient's manufacturers (Fonterra and Danisco for the first, DuPont for the second, Danisco China for the third)17,18,19. The finished 12-strain WonderBiotics formula hasn't been tested as a whole; the evidence above applies to HN019 as an ingredient, and the matrix pairs this product with a fiber option for readers whose main complaint is hard, infrequent stools.
Align's single strain, B. longum 35624, has two randomized trials behind it from overlapping, industry-linked investigator groups (both include O'Mahony, Kiely, and Shanahan as authors), each showing benefit on bloating and abdominal pain or discomfort at a different dose: a 362-woman trial, sponsored by Procter & Gamble's Health Sciences Institute with at least one P&G-affiliated author, found the mid-dose (10⁸ CFU) beat placebo and both other doses on its primary pain-and-discomfort endpoint20; a smaller 77-person trial found benefit at a higher dose (10¹⁰ CFU) on bloating, distension, and pain across most treatment weeks21. Neither trial's dose matches the current label's guaranteed shelf-life potency of 10 million CFU exactly. Neither trial enrolled GLP-1 users or was designed around constipation.
Seed's DS-01 has a published, peer-reviewed, placebo-controlled trial on a finished, multi-strain formula for bloating and gas in generally healthy adults who weren't selected for GLP-1 use: 350 adults with self-reported bloating and digestive discomfort were randomized, with 219 providing evaluable data at week 6 (the primary analysis required only one adherent post-baseline week, and the final two timepoints used last-observation-carried-forward for missing data). The full 24-strain, 53.6-billion-AFU formula significantly improved a GI quality-of-life score (primary endpoint, P<0.05) and a combined bloating-and-gas score (secondary endpoint, P<0.01) over 6 weeks22. Seed Health funded the study through a grant; trial operations were performed independently by a third-party research operator. Two authors are Seed employees or shareholders, and two more, including one who is separately an employee of Increnovo, serve as advisors to Seed22.
Culturelle Digestive Daily's label sticks to occasional gas, bloating, and diarrhea and doesn't claim to help constipation, which matters if regularity is your main goal. Fiber
WonderBiotics Daily Fiber Mix combines Fibersol (a soluble corn fiber chemically related to, but not the same tested product as, the wheat dextrin below), PHGG, and psyllium with FOS and polydextrose in one 6 g sachet. Lab fermentation testing of wheat dextrin, PHGG, and inulin (not Fibersol, psyllium, or FOS) found PHGG ferments more slowly and produces less gas than inulin, while wheat dextrin produced the least gas of the three23. Psyllium is separately classified as nonfermentable rather than slow-fermenting, a different mechanism for tolerability24. FOS belongs to the fructan family, which tends to ferment quickly once it reaches the colon. The blend mixes fibers with different fermentation profiles rather than being uniformly slow, consistent with the label's own guidance to expect some adjustment gas in the first week or two and to start at one sachet a day.
Psyllium, the fiber in Metamucil and ColonBroom, has the strongest bowel-frequency data of any fiber type here: a 2022 systematic review and meta-analysis of trials in adults with chronic constipation found fiber overall raised weekly bowel movements meaningfully, and psyllium showed an even larger increase in its own subgroup, though results varied a lot from trial to trial24. Those psyllium trials generally used more psyllium per day than a single Metamucil packet provides, and the same review found that fiber's frequency benefit held up mainly at the higher end of the doses tested24, worth knowing since one packet falls well under that range. The review also found a significant increase in flatulence severity across fiber types, based on a small number of trials24, a signal that gas can increase with fiber supplementation without proving every fiber type here raises gas equally.
A 2013 in vitro fermentation study, funded by Novartis Consumer Health (Benefiber's manufacturer at the time, with a co-author who was a Novartis employee), found wheat dextrin produced the least gas of the fibers it tested in a lab fermentation vessel23. That's a lab measurement of gas production, not a human bloating trial; standalone human evidence on wheat dextrin's effect on bowel frequency is thinner than psyllium's. A 2022 randomized trial of PHGG, the fiber WonderBiotics' blend also uses, in 52 long-term care residents found no significant difference in bowel frequency versus a water-only control; laxative use decreased significantly at weeks 3 and 425.
Supergut GLP-1 Daily Support markets a 6-to-7-gram stick pack of oat beta-glucan, resistant potato starch, and green banana starch with mechanism-based language about supporting the body's own GLP-1 signaling. The company's own published trial tested a different product: a 15-gram-fiber, 15-gram-protein meal-replacement shake in people with type 2 diabetes, which improved a diabetes-distress score and HbA1c but never directly measured GLP-1 itself25. That trial doesn't transfer cleanly to the lower-dose stick pack sold today. PGX Daily's glucomannan and Metamucil or ColonBroom's psyllium both work by forming a viscous gel in the gut, which is why their labels require gradual titration and a full glass of water with every dose; a 1986 case report tied glucomannan tablets taken without enough water to esophageal obstruction26.
Fiber and GLP-1 Safety Considerations
GLP-1 drugs slow gastric emptying and, based on their receptor distribution throughout the gut, are thought to reduce peristalsis and alter the migrating motor complex that regulates fasting gut motility. A 2025 case report described a woman with a prior history of small bowel obstruction who developed recurrent obstruction after starting semaglutide; the authors reported a temporal association and discussed GLP-1 receptor activation as a possible contributor, not as an established cause27. Separately, at the population level, the evidence on GLP-1 drugs and intestinal obstruction is mixed: one study found a modestly higher hospitalization rate for obstruction versus another diabetes drug class (HR 1.69)28, while a larger, more recent Scandinavian cohort study found no confirmed increase in risk29. Neither of those two cohort studies measured gastric emptying or gut motility directly; they looked at hospitalization and diagnosis rates.
Use viscous, gel-forming fibers, psyllium and glucomannan specifically, with a full glass of water every time, start at the label's lowest dose, and add only one new probiotic or fiber product at a time so you can tell what's causing what. PHGG and Fibersol don't form the same kind of viscous gel and aren't tied to the same hydration warnings. Severe abdominal pain, persistent vomiting, or an inability to pass gas or stool at all are signs of a possible bowel obstruction, not ordinary constipation; Mayo Clinic lists those together as reasons to seek care right away rather than adding another supplement30.
How to Use the Matrix
