The right answer depends on your symptom. WonderBiotics Probiotics for Gut Health targets constipation and regularity with published strain-level data; Align Probiotic has the longest track record for bloating and gas; Seed DS-01 has the strongest trial on its complete formula; Ritual and Culturelle are simpler, lower-commitment options for occasional digestive upset on semaglutide.
Why Semaglutide Changes What You Need From a Probiotic
GLP-1 medications slow gastric emptying, which is part of how they work, and part of why so many users end up with new or worse digestive symptoms. If constipation is the symptom semaglutide gave you, WonderBiotics Probiotics for Gut Health is built around this specifically. It's a 60-billion-CFU, 12-strain blend,14 and its regularity claims trace back to Bifidobacterium lactis HN019, one of the few probiotic strains with published human data on bowel movements rather than just marketing language.
In a 100-person dose-ranging trial funded by Fonterra, the high-dose HN019 group's whole-gut transit time fell from an average of 49 hours to 21 hours over the study period, a change of about 28 hours. The low-dose group improved by about 18.5 hours, and the placebo group barely moved (+1.3 hours), with the difference across all three arms statistically significant (P<0.001).6 In a separate 228-person trial funded by DuPont, HN019's manufacturer at the time, HN019 didn't beat placebo on its pre-specified primary or secondary endpoints overall.7 Only in a post-hoc subgroup of 65 people who started with three or fewer bowel movements a week did combined-dose HN019 improve weekly frequency (P=0.01), with straining relief limited to the high-dose group.7 A more recent 229-person trial in adults with functional constipation, using the same strain at a comparable dose, found HN019 didn't outperform placebo on its pre-specified primary endpoint either (weekly complete spontaneous bowel movements, intention-to-treat P=0.37).16 Taken together, HN019's evidence is a mix of one positive dose-response trial on transit time, one subgroup-only result inside an otherwise negative trial, and one more recent negative primary-endpoint trial. None of the three enrolled anyone on a GLP-1 medication.

Those are our marketing claims about the finished formula, not numbers from an independent trial, and the same gap applies to Ritual's and Culturelle's blends, since none of the three has run a full-formula RCT. Seed's DS-01 doesn't market specifically for constipation, but its finished-formula trial did report a significant secondary improvement in constipation symptoms and self-reported regularity versus placebo, discussed below.3 WonderBiotics is the only one of the five built specifically around constipation and regularity as its primary claim, and the strain behind that claim has the most direct, if mixed, bowel-frequency evidence of the group.
Best for Bloating and Gas: What the Trials Actually Show
For bloating specifically, Seed currently has the strongest trial behind any single brand on this list. In a 2026 randomized, double-blind, placebo-controlled trial, 350 adults with self-reported bloating and indigestion were randomized to the complete DS-01 formula or placebo; 219 supplied evaluable data at the Week 6 primary time point, with missing values carried forward from participants' last observation. On that basis, the formula improved the pre-specified primary endpoint (GI-related quality of life) versus placebo (P<0.05), and the secondary bloating/gas score also improved (P<0.01), with 72.3% of the synbiotic group reporting "never/rarely" bloating versus 55.9% on placebo (P<0.001).3 Seed Health funded the trial; two authors are Seed employees or shareholders who contributed to the study's conceptualization and formal analysis, and two more serve as paid advisors to Seed. Day-to-day clinical operations were run by an outside contract research organization, Radicle Science.3 It's still the only trial on this list testing a finished, marketed formula rather than an individual strain in isolation.
Align's evidence is older and strain-specific rather than formula-specific, but it's substantial. In a 362-woman dose-ranging trial funded by Procter & Gamble, the middle dose of B. infantis 35624 (now reclassified as B. longum 356249) beat placebo on the primary endpoint of abdominal pain/discomfort, with global symptom improvement exceeding placebo by more than 20% (P<0.02); bloating improved too, though that reading sits below the primary endpoint in the evidence hierarchy.4 A separate 77-person trial found the same strain, at a different dose, produced significantly lower bloating/distention scores than placebo on most of the individual weekly symptom measures tracked over 8 weeks; its funding wasn't disclosed in the sections available for review.5 The two trials tested doses of 108 and 10¹⁰ CFU, and Align's commercial capsule is labeled at 10⁹ CFU at manufacture (guaranteed only down to 10⁷ CFU by the end of shelf life),13 in between the two trial doses rather than an exact match to either.
Ritual's Synbiotic+ pairs LGG and BB-12, two individually well-studied strains, with a bacteriophage-based prebiotic (PreforPro) and a tributyrin postbiotic; the brand advertises "100+ publications" behind the two probiotic strains generally, but that's literature on LGG and BB-12 across many products and doses, not a trial of this specific four-ingredient combination.10 Culturelle's gas/bloating claim rests on the same kind of strain-level evidence as Ritual's, via LGG and B. lactis CECT 8145, with no finished-formula trial of its own.8 WonderBiotics' bloating claim carries the same caveat noted above.

Ritual (2 strains plus a prebiotic and a postbiotic), WonderBiotics (12 strains plus FOS and an enzyme blend), and Seed (24 strains plus a polyphenol extract) are all more complex formulas. More strains and added prebiotic fiber aren't inherently harder on digestion, but WonderBiotics' product page doesn't break out the individual HN019 or FOS quantities inside its 60-billion-CFU blend, so there's no way to compare its actual per-strain dose against the HN019 trials above.14 A more crowded label does mean more variables if something doesn't agree with you, which is part of why we recommend starting at a lower intake and watching your own response for the first couple of weeks rather than assuming a bigger number on the label means more relief, faster.

WonderBiotics, Culturelle, and Align all name every strain in their formula on the product label or page. Ritual names both of its two strains. Seed's main DS-01 product page groups its 24 strains into four functional categories (digestive, dermatological, cardiovascular, micronutrient) by AFU count rather than naming them there.11 The individual species and strain codes for all 24 are published on a separate Seed reference page,17 so the full list is available on seed.com, just not on the page most people order from. Seed currently has the best finished-formula trial data on this list; finding its complete strain breakdown takes an extra step that the other four brands don't require.
What Nobody Can Claim Yet: Direct Evidence in GLP-1 Users
None of the trials cited above, across any of the five brands, enrolled people taking semaglutide, tirzepatide, or any other GLP-1-based medication. The strain and formula research behind HN019, B. longum 35624, LGG, BB-12, and Seed's DS-01 blend was done in general adult, IBS, or healthy-volunteer populations. A GLP-1-specific trial is registered (NCT07213323) but had not started recruiting as of 2026.2 Brand language that frames a probiotic as a "GLP-1 companion" or built to ease medication side effects — including WonderBiotics' own — is a positioning claim based on how the formula was designed, not a result from testing people actually on the drug.
The absence of GLP-1-specific trials is a reason to treat brand-specific superiority claims skeptically across the board and to judge each product on the strain-level evidence that does exist rather than on marketing percentages attached to the finished formula.
How to Introduce a Probiotic Safely While on Semaglutide
Start with one product at a time and the lower end of the labeled dose, so if something changes in your gut you can tell what caused it. Mild, temporary gas or bloating in the first stretch is a commonly reported adjustment response when starting a new probiotic, though how long it takes to settle varies by product and person; give it a couple of weeks before deciding whether it's working, on or off a GLP-1 medication.
Talk to your prescriber or pharmacist before adding any new supplement, especially during a dose increase, since that's when GI side effects are most likely to spike regardless of what else you're taking. A probiotic is a supportive add-on; adjusting your dose or diet with your care team is what actually addresses semaglutide's GI side effects.
Know the difference between ordinary adjustment symptoms and a red flag. If bloating and gas are the bigger issue and you want the single strongest trial behind a finished product, Seed DS-01 currently has that, though its full strain list sits on a separate reference page rather than the product page itself. If you want the simplest possible formula to rule in or out as the cause of a new symptom, Align is the one-strain option. If budget is the deciding factor and your symptoms are mild and occasional, Culturelle is the least expensive way to try a two-strain, well-studied combination. Ritual sits in the middle: a simple two-strain base with added prebiotic and postbiotic ingredients, at a mid-range price.
